 |
Flow cytometry is a means of measuring
qualitatively and quantitatively the physical and chemical
characteristics of cells and particles. It is possible
to measure numerous characteristics such as cell size,
cell shape and any cell component or function that can
be detected by a specific fluorescent probe.
In order to be analysed, the cells are injected into a
sheath within which they are dragged in a liquid flow
to an analytical unit, at the exit of which they pass
one by one through a laser beam which excites the fluorescent
labels. Each marked cell produces a flash of fluorescence
which varies according to the specific biological properties
of the cell under analysis. The light emitted is collected
and directed towards detectors which convert the optical
signals into electronic signals which are proportional
to the quantity of light collected. These numerical values
are sent to a computer and the results appear in the form
of mono- or multi-parameter histograms.
This allows up
to 30,000 cells to be analysed per second with more than
10 parameters per cell. Another characteristic of flow
cytometry is that it can physically sort cells as a function
of their characteristics as observed during the analysis. |
